Abstract

Introduction: The overall haemostatic potential (OHP) assay was applied for testing of anticoagulant action of sodium salt of calix[4]arenemethylene-bis-phosphonic acid (C-145) in blood plasma In vitro and In vivo.

Methods: Calix[4]arene C-145 was added to platelet poor plasma or injected intravenously to rabbits in equal concentrations (20 µM), clotting was initiated by APTT-reagent and turbidity change of the mixture was measured in the presence or absence of tPA.

Results: We demonstrated that in vitro 20 μM of C-145 substantially decreased the OHP in 1.5 times and did not influence fibrinolysis. That injection of calix[4]arene in vivo decreased the overall haemostatic potential in 30-10%. The prolongation of half-time of clot lysis was not observed.

Conclusion: Overall haemostatic potential assay allowed us to conclude that coagulation and fibrinolytic branches of haemostasis remain in dynamic balance at C-145 administration. C-145 being taken in the final concentration 20 μM inhibited coagulation potential of blood plasma and did not affect fibrinolysis in vivo.
