Abstract

Ganoderma is a traditional medical fungus and its complex species bear highly similar morphological features. Thus, identification of the Ganoderma species relied heavily on taxonomic remains unclear. In this study, the basidiocarp (fruiting body) morphology combined with internal transcribed spacer (ITS) sequences, phylogenetic, aligned rDNA sequence, and secondary metabolite profile were used to identify Ganoderma species in Viet Nam. According to basidiocarp morphology, the GLVN02 and GLVN05 were predicted as Ganoderma species. By determining the ITS1-phylogenic of eight species clearly identified GLVN02 is a Ganoderma lucidum. GLVN05 is a subgroup of Ganoderma tsugae species and indicated their homology formed another subgroup, whereas the other species were unidentified. Aligned ITS1-rDNA sequence of GLVN02 and GLVN05 were similarly 100% nucleotides to Ganoderma lucidum (MF476197 and JQ520168) and Ganoderma tsugae (JQ520215) on Genbank. Further, GLVN02 and GLVN05 have only T/C substitution, in fact, several variable nucleotide discrepancies A/G, T/C or T/G substitutions frequently occurred and differentiated to GLVN01, GLVN03, GLVN04, GLVN06, GLVN07, and GLVN08. Secondary metabolite extracted from basidiocarp of GLVN02 contains much more polysaccharide than the others. These results indicate that the phylogenetic validation ITS1 sequence and provide updated taxonomic and diverse of Ganoderma species in Viet Nam.
