Abstract

Background and Objectives: The oyster mushroom (Pleurotus florida) is an edible mushroom in both forms fresh as well as dry. It has been perceived as an exceptionally potential converter of agricultural waste into profitable protein. The present investigations were carried out to find the suitable cereal grains extract media for mycelial extension of oyster mushroom (Pleurotus florida). Materials and Methods: The present work was conducted at Lovely Professional University during 2017-18. For the evaluation of mycelia extension of oyster mushroom (Pleurotus florida) seven different media viz. wheat dextrose agar, wheat extract agar, sorghum dextrose agar, sorghum extract agar, maize dextrose agar, maize extract agar and pearl millet dextrose agar were taken. Thereafter consequences of pH (in the range of 5.0-8.0) and temperature (15, 20, 25, 30, 35 and 40°C) on the growth of Pleurotus florida was investigated.
Results: Among all tested media, the significant superior mycelial extension was observed on pearl millet dextrose agar media (76 mm) followed by wheat extract agar (70 mm) and at pH 6 (77.33 mm) nearly followed by pH 6.5 (74.67 mm) after 6 days incubation at 25±2°C. In all used temperatures, generally, average maximum linear growth was observed at a temperature range 25°C (77.73mm) followed by 20°C (64mm). Conclusion: Therefore, it can be concluded that pearl millet dextrose agar media, pH 6 and temperature level 25°C supported the best mycelial growth of oyster mushroom (Pleurotus florida). This study will be very useful for maintaining the pure culture of Pleurotus florida.
