Abstract

Background: It is well known that diabetes mellitus type 2 is a multi-factorial disease involving multiple genes and environmental factors to varying extent. In present investigation we are trying to investigate role of gene polymorphism in ABCA1 gene and tsh level with diabetes type 2.

Objective: In present study we aimed to investigate the relationship between ABCA1 C69T (rs18000977) gene polymorphism and serum TSH level in susceptibility to diabetes type 2.

Materials and Methods: 590 unrelated subjects comprising to 280 T2D patients and 310 ethnically matched controls of central Indian population recruited in this study. Anthropometric and life style data were collected during sampling. The C69T polymorphism of ABCA1 gene has been amplified by PCR-RFLP analysis. Genotype allele frequency and carriage rate of ABCA1 gene polymorphism were recorded. Serum TSH level is examine by assay of TSH (based upon sandwich ELISA method).

Results: Diabetic patients had markedly higher levels of fasting plasma glucose (P<0.0001) Postprandial glucose (P<0.0001) and HbA1c (P<0.0001) and triglyceride (P<0.0001) compared to that of control subject. The genotype distribution of ABCA1 was significantly different in control as compared to diabetic patients. The frequency of CC genotype (0.01140)* and C allele (0.0046)** was significantly higher in patients thane controls and associated with higher risk. An odds ratio of 1.442 of ‘C’ allele showed stronger association of overall “C” allele frequency in diabetes susceptibility. Serum TSH level slightly higher in healthy control group as compare to diabetic group (2.9±0.85 in control vs 2.78±1.21 in case) although difference was nominal and statistically non-significant (P value 0.1608).

Conclusion: Our study indicates that ABCA1 gene polymorphism has a strong association with diabetes type 2 but serum TSH level lacks any association with diabetes type 2 in central Indian population.
